Dengue virus (DENV) is an enveloped, single-stranded, positive-sense RNA virus that includes four related but distinct serotypes (DENV1, 2, 3, and 4). It encodes three structural proteins and seven non-structural proteins including NS1, a highly conserved glycoprotein which is essential for viral replication and viability.
During virus infection, NS1 is found associated with intracellular organelles or is alternatively transported through the secretory pathway to the cell surface. Since the NS1 glycoprotein is secreted into the bloodstream, it can be used as a marker in diagnostic for early detection of dengue infection.
A2311 is composed of the full length amino acid sequence of the NS1 antigen of dengue virus type 1 which was cloned into an insect expression vector. The secreted and soluble recombinant protein was purified with affinity chromatography.
